アレーンおよびヘテロアレーンは、典型的には、有機リチウム化合物または有機リチウムアミドを用いた指向性リチオ化により脱プロトン化される（例えば、有機リチウムとしてLDAが使用される）。これらの試薬の高い反応性と求核性は、しばしば望ましくない副反応を引き起こし、エステルやケトンなどの敏感な官能基の存在を誘発する。さらに、このような脱プロトン化反応では低温が必要になることが多く、大規模になると生産コストが高くなる。
亜鉛アミドは、その低い運動塩基性度により、多くの官能基を許容する。
